Many clients reported similar issues with packaging and Datasmith.

Client report that It's a project that they've been working on since 4.19. It was imported using Datasmith 4.19. It used to package without problems on UE 4.20.3. Now this is their first attempt to package this project/level after updating to 4.21.1, and I got this error.

Repro Step
1. Extract the following plugin to your UE_4.21/Engine/Plugins/Marketplace folder.

2. Create a new project (Unreal Studio / Blank), activate the attached plugin and the Substance plugin, close the editor.
3. Extract this file to the project's Content folder:

4. Open Unreal, set the game default map to Maps/HLar and add it to the list of maps, set package mode to Shipping, and attempt to package the game.
